目的对天麻种子的抗抑郁作用进行研究,为其开发利用提供依据。方法将小鼠随机分为空白对照组、模型组、盐酸氟西汀组及天麻种子高、中、低剂量组,连续灌胃给药28 d。通过小鼠强迫游泳和悬尾抑郁模型,观察小鼠4 min内游泳不动时间和悬尾不动时间;在悬尾实验结束24 h后,每只小鼠首先进行5 min悬尾实验再进行5 min强迫游泳实验。30 min后摘眼球取血,测定小鼠血清中促肾上腺皮质激素释放激素(CRH)、肾上腺皮质激素(ACTH)、皮质酮(CORT)含量。结果与模型组比较,各给药组小鼠强迫游泳不动时间和悬尾不动时间均显著减少(P<0.05,P<0.01);与模型组比较,天麻种子组均可降低小鼠血清中CRH、ACTH和CORT含量,天麻种子高、中剂量组效果显著(P<0.05,P<0.01),并且天麻种子高剂量组小鼠血清激素水平接近盐酸氟西汀组。结论天麻种子具有抗抑郁作用,其作用机制可能与调节HPA轴激素水平有关。
Objective To study the antidepressant effects of Gastrodia elata Seeds and provide the basis for its development and utilization. Methods The mice were randomly divided into blank control group, model group, fluoxetine hydrochloride group and high, medium and low dose groups of Gastrodia elata Seeding. After forced swimming and tail-suspended depression model mice were observed within 4 min swimming immobile immortality and tail immobile time; 24 h after the tail suspension experiment, each mouse first 5 min tail test again 5 min forced swimming test. After 30 min, the eyeballs were taken for blood and the content of corticotropin-releasing hormone (CRH), adrenocorticotropic hormone (ACTH) and corticosterone (CORT) in the serum of the mice were measured. Results Compared with the model group, the forced swimming time and suspended tail immobilization time in each group were significantly decreased (P <0.05, P <0.01). Compared with the model group, The contents of CRH, ACTH and CORT, the effect of high and medium dose of Gastrodia elata seeds were significant (P <0.05, P <0.01), and the level of serum hormone in the gastrodia tuber high dose group was close to fluoxetine hydrochloride group. Conclusion Gastrodia elata indica has antidepressant effect and its mechanism may be related to the regulation of HPA axis hormone levels.
