目的分析钾通道KCNQ1在不同发育阶段大鼠胰腺以及成年大鼠胰岛中的表达情况,为探悉其在胰腺B细胞发育及功能中可能的作用提供实验依据。方法采用高密度寡核苷酸芯片对胚胎第12.5天(E12.5)、E15.5、E18.5、新生和成年大鼠胰腺进行基因转录水平分析,并用RT-PCR对KCNQ1通道亚基在上述5个时期大鼠胰腺以及成年大鼠胰岛中的表达进行验证和分析。结果与胰腺内分泌细胞分化、成熟有关的转录因子分别于E15.5、E18.5达到表达高峰;E18.5胰腺中B细胞功能成熟标志基因的表达显著增加,而KCNQ1钾离子通道的编码基因KCNQ1及KCNE1此时才出现表达。成年胰腺及胰岛中均具有多种KCNQ1通道亚基的表达。结论 KCNQ1通道的表达出现于胰腺发育后期内分泌细胞功能逐渐成熟阶段,在成年胰岛中亦具有较高表达水平,提示KCNQ1通道可能表达于成熟B细胞并参与其内分泌功能。
Objective To analyze the expression of potassium channel KCNQ1 in rat pancreas and adult rat islets at different developmental stages, and provide experimental evidence for its possible role in the development and function of pancreatic B cells. Methods High-density oligonucleotide microarray was used to analyze the gene transcription level of the pancreas on embryonic day 12.5 (E12.5), E15.5, E18.5, newborn and adult rats. RT-PCR was used to detect the expression of KCNQ1 channel subunit The above five periods of rat pancreas and adult rat islets in the expression of verification and analysis. Results The transcriptional factors related to the differentiation and maturation of pancreatic endocrine cells reached their peak at E15.5 and E18.5, respectively. The expression of functional maturation markers of B cells was significantly increased in E18.5 pancreas, while the expression of KCNQ1 And KCNE1 only appeared at this time. There are many KCNQ1 channel subunits in adult pancreas and islets. Conclusions The expression of KCNQ1 channel appears in the stage of gradual maturation of endocrine cells in the late stage of pancreas development, and also has a higher expression level in adult pancreatic islets, suggesting that KCNQ1 channel may express in mature B cells and participate in its endocrine function.
