目的 :建立携带示踪标记的永生化大鼠肝细胞系。方法 :用脂质体转染法将增强型绿荧光蛋白 (EGFP)基因转入永生化大鼠肝细胞中。结果 :转染 3d后 ,在荧光显微镜下观察 ,对照组肝细胞不发荧光 ;实验组肝细胞发绿色荧光 ,转染率为 4 1% ,经G4 18筛选后得到稳定表达的肝细胞。结论 :应用基因转染技术将EGFP基因转入永生化大鼠肝细胞中并获得稳定表达。
OBJECTIVE: To establish an immortalized rat hepatocyte line carrying a tracer marker. Methods: The green fluorescent protein (EGFP) gene was transfected into immortalized rat hepatocytes by lipofection. Results: After transfection for 3 days, the hepatocytes in the control group showed no fluorescence under the fluorescence microscope. The hepatocytes in the experimental group developed green fluorescence with a transfection rate of 41%. The stable hepatocytes were obtained after being screened by G418. Conclusion: EGFP gene was transfected into immortalized rat hepatocytes by gene transfection technique and stably expressed.
