应用RAPD技术研究闽香鳢及其亲本之间在遗传结构、种群多样性上的相互关系,由此为探讨种质鉴定提供依据。在所用的90个随机引物中,有35个引物在闽香鳢及其亲本之间呈现多态。通过RAPD分析,获取了闽香鳢及其亲本的RAPD指纹图带。结果表明闽香鳢与斑鳢或乌鳢的遗传距离要比其父母本之间的遗传距离近,即闽香鳢为父母本杂交所产生的后代。闽香鳢种群的多态性比例47.9%与杂合度H=0.122,可作为闽香鳢遗传育种的指标来检测后代的群体遗传多样性。通过多态性比较,获得2条具有群体差异性的DNA片断,大小分别为910 bp和1 011 bp,经过测序后合成特异性引物,此引物可特异性区分上述3种鱼。
RAPD technique was used to study the relationship between genetic structure and population diversity of Min Hong 鳢 and its parents, thus providing evidence for germplasm identification. Of the 90 random primers used, 35 primers showed polymorphism between Min Hong and its parents. By RAPD analysis, we obtained the RAPD fingerprinting bands of Min Hong 鳢 and its parents. The results showed that the genetic distance between Min Hong 鳢 and Cantaris or Malus sibiricus was closer than the genetic distance between their parents, that is, the offspring of Min Hong 鳢 as parents hybrid. The percentage of polymorphism in the population of Min Hong tree was 47.9% and its heterozygosity was 0.122. It could be used as an index of genetic breeding of Min Hong tree to detect the population genetic diversity of offspring. Two DNA fragments with population diversity of 910 bp and 1 011 bp were obtained by comparison of polymorphisms. After sequencing, specific primers were synthesized, which could specifically distinguish the three kinds of fish.
