目的:研究人类精浆中一类特异性的抗原成份(精子外套抗原sperm-coatingantigens,SCA)。方法:本实验用已制备得的一株抗人精浆来源的精子外套抗原单克隆杂交瘤细胞株D1.在小鼠腹腔增殖生产大量腹水单克隆抗体,通过快速蛋白液相层析仪(FPLC)系统的羟基磷灰石柱,一次性从小鼠腹水中纯化此抗体,并制备免疫亲和层析柱,从精浆中分离到相应的抗原成份。结果:经鉴定该抗原成份分子量为20kD,来源于精囊腺,具有器官特异性。结论:该SCA的提纯,为抗精子避孕疫苗的研制提供了基础实验材料,并且有助于进一步研究其在生殖活动和免疫不孕病因中的作用。
Objective: To study a class of specific antigens in human seminal plasma (sperm-coating antigens, SCA). Methods: In this experiment, a monoclonal anti-human seminal plasma antigen-derived monoclonal hybridoma cell line D1 was prepared. A large number of ascites monoclonal antibodies were produced by peritoneal proliferation in mice and the antibody was purified from the ascites of mice once by the hydroxyapatite column of the fast protein liquid chromatography (FPLC) system and the immunoaffinity chromatography column was prepared. From the seminal plasma isolated corresponding antigen components. Results: The molecular weight of the antigen was identified as 20kD, derived from the seminal vesicle and organ-specific. Conclusion: The purification of SCA provides the basic experimental material for the development of anti-sperm contraception vaccine, and helps to further study its role in the causes of reproductive activities and immune infertility.
