The first report of the use of familial adenomatous polyposis (FAP) gene-associated DNA probes estimated the risk of asymptomatic persons suffering from the disease. A total of seven asymptomatic individuals (ages 0-39 years) with FAP families who were at risk for the disease were studied. In each case, six DNA probes were used to detect genotypes. The DNA purified from peripheral blood leukocytes is first digested with appropriate restriction enzymes, cut into specific fragments by agarose gel electrophoresis, and hybridized with radiolabeled DNA probes. Then autoradiograph display supplemented with this probe at -70°C for 24-48 hours
