目的:研究清润方对2型糖尿病(type 2 diabetes mellitus,T2DM)大鼠氧化应激及炎症反应的影响。方法:采用尾静脉注射链脲佐菌素(streptozotocin,STZ)加高糖高脂饲料饲养建立大鼠T2DM模型,将大鼠随机分为空白对照组、模型组、清润方大、中、小剂量组(0.5,1,2 g.kg-1.d-1)、二甲双胍组(150 mg.kg-1.d-1),各治疗组分别ig给予相应药物治疗。干预治疗4周后检测各组大鼠空腹血糖(FBG)、血清胰岛素水平;肝脏组织丙二醛(MDA)含量、超氧化物歧化酶(SOD)活性;血清白细胞介素6(IL-6)、肿瘤坏死因子α(TNF-α)含量,观察清润方对糖尿病大鼠氧化应激及炎症反应的影响。结果:与模型组比较,清润方各剂量组TNF-α水平降低(P<0.05),中、小剂量组IL-6水平、肝脏MDA含量降低(P<0.05);中、大剂量组肝脏组织的SOD活性提高(P<0.05)。结论:清润方对糖尿病大鼠的抗氧化应激及减轻炎症反应作用可能是其发挥治疗作用的机制。
Objective: To investigate the effect of Qingrun formula on oxidative stress and inflammation in type 2 diabetes mellitus (T2DM) rats. Methods: T2DM model was established by injecting streptozotocin (STZ) and high glucose and high fat diet in the tail vein. The rats were randomly divided into blank control group, model group, Qingrun Fang large, medium and small dose (0.5,1,2 g.kg-1.d-1), metformin group (150 mg.kg-1.d-1). The treatment groups were given the appropriate drug therapy. After intervention for 4 weeks, the levels of fasting blood glucose (FBG), serum insulin, the contents of malondialdehyde (MDA) and superoxide dismutase (SOD) in liver tissue were measured; the levels of serum interleukin 6 (IL- , Tumor necrosis factor α (TNF-α) in diabetic rats to observe the effects of Qingrun formula on oxidative stress and inflammatory response in diabetic rats. Results: Compared with the model group, the levels of IL-6 in the middle and low dose groups and the content of MDA in the liver decreased (P <0.05) SOD activity increased (P <0.05). Conclusion: The effect of Qingrunfang on anti-oxidative stress and anti-inflammation in diabetic rats may be the mechanism of its therapeutic effect.
