目的:观察灵芪胶囊(LQC)体外对人肺腺癌A549细胞增殖及凋亡相关基因表达的影响。方法:体外人肺腺癌A549细胞经LQC含药血清处理后,采用MTT比色法测定其抑制率;逆转录-聚合酶链反应检测其凋亡相关基因bcl-2、bax的mRNA表达;Wright-Giemsa染色法观察其形态变化。结果:不同浓度LQC含药血清对A549细胞增殖具有抑制作用,且与浓度呈正相关;LQC含药血清能使bcl-2基因的mRNA表达水平显著下降、bax基因的mRNA表达水平显著增强;同时,细胞形态发生改变。结论:LQC含药血清可调控肿瘤细胞凋亡的相关基因,这可能是其产生抗肿瘤生物学效应的基础之一。
Objective: To observe the effect of Lingqi Capsule (LQC) on proliferation and apoptosis-related gene expression of human lung adenocarcinoma A549 cells in vitro. Methods: Human lung adenocarcinoma A549 cells were treated with LQC-containing serum in vitro and the inhibitory rate was determined by MTT colorimetric assay. The mRNA expression of apoptosis-related genes bcl-2 and bax was detected by reverse transcription-polymerase chain reaction. Wright -Giemsa staining observed morphological changes. Results: The different concentrations of LQC-containing serum could inhibit the proliferation of A549 cells and had a positive correlation with the concentration of LQC-containing serum. The expression of bcl-2 mRNA and the mRNA of bax gene were significantly increased in the LQC- Cell morphology changes. Conclusion: LQC-containing serum can regulate tumor cell apoptosis-related genes, which may be the basis of its anti-tumor biological effects.
