目的:研究口腔黏膜癌前病变癌变过程中Stat3、Bcl-2及cyclinD1的表达及其相关性。方法:采用免疫组化的方法分别检测正常口腔黏膜、单纯性增生黏膜、异常增生黏膜与口腔鳞状细胞癌(oral squamous cell carcinoma,OSCC)中Stat3、Bcl-2及cyclinD1的表达。结果:正常口腔黏膜、单纯性增生黏膜、异常增生黏膜和OSCC中Stat3和Bcl-2、cyclinD1的积分光密度值(intergrated optical density,IOD)均逐步升高;Stat3、Bcl-2、cyclinD1三者的IOD值除单纯性增生黏膜与口腔正常黏膜无统计学意义外,其余各组间均有统计学意义。Stat3与Bcl-2 IOD值呈线形正相关;Stat3与cyclinD1 IOD值呈线形正相关;Bcl-2与cy-clinD1 IOD值无相关性。结论:Stat3、Bcl-2及cyclinD1在口腔黏膜癌前病变癌变中表达的相关性提示,口腔黏膜癌前病变癌变是由多种基因共同参与的过程。
OBJECTIVE: To investigate the expression of Stat3, Bcl-2 and cyclinD1 in carcinogenesis of oral precancerous lesions and their correlation. Methods: The expressions of Stat3, Bcl-2 and cyclinD1 in normal oral mucosa, simple proliferative mucosa, dysplastic mucosa and oral squamous cell carcinoma (OSCC) were detected by immunohistochemistry. Results: The values of Stat3, Bcl-2 and intergrated optical density (IOD) of cyclinD1 in normal oral mucosa, simple hyperplastic mucosa, dysplastic mucosa and OSCC all increased gradually. Stat3, Bcl-2 and cyclinD1 IOD values in addition to simple hyperplastic mucosa and oral mucosa was not statistically significant, the rest of the groups were statistically significant. There was a linear positive correlation between Stat3 and Bcl-2 IOD, a positive linear correlation between Stat3 and cyclinD1 IOD, and no correlation between Bcl-2 and cy-clinD1 IOD. Conclusion: The correlation of Stat3, Bcl-2 and cyclinD1 expression in carcinogenesis of oral mucosa precancerous lesions suggests that the carcinogenesis of oral mucosa precancerous lesions is a process involving multiple genes involved.
