目的:半边旗的植化研究及其有效部位和有效成分的抗肿瘤活性研究。方法:半边旗乙醇提取物上活性炭柱,乙醇洗脱液减压浓缩得粗提物(C),甲醇氯仿(2∶1)洗脱液浓缩后以硅胶柱层析分离,得到一个倍半萜(B),两个二萜(4F、5F)。B、4F、5F的结构经光谱鉴定,尤其是用二维NMR确证了空间结构。以台盼蓝染色作活细胞计数,算出药物对细胞的生长抑制率,IC50按改良Karber公式计算。结果:分离得到三个化合物,确定了结构。以HL60细胞为筛选模型,结果表明:粗提物(C)、化合物5F明显抑制HL60细胞增值。结论:药理实验表明,粗提物的抗肿瘤活性明显高于5F,预示粗提物中仍有高活性化合物,值得进一步研究。
OBJECTIVE: To study the phytochemicals of Pteris semipinnata L. and its anti-tumor activity of its effective sites and active ingredients. Methods: The ethanol extracts of Pteris semipinnata L. on activated charcoal column were concentrated under reduced pressure with ethanol eluent to obtain crude extract (C). The methanol-chloroform (2:1) eluate was concentrated and separated by silica gel column chromatography to obtain a half-fold.萜 (B), two two 萜 (4F, 5F). The structures of B, 4F and 5F were identified by spectra, and the spatial structure was confirmed by two-dimensional NMR. Trypan blue staining was used as the viable cell count to calculate the growth inhibition rate of the drug on the cells. IC50 was calculated according to the modified Karber’s formula. Results: Three compounds were isolated and the structure was determined. Using HL-60 cells as a screening model, the results showed that the crude extract (C) and compound 5F significantly inhibited the proliferation of HL-60 cells. Conclusion: Pharmacological experiments showed that the anti-tumor activity of the crude extract was significantly higher than 5F, suggesting that there are still high active compounds in the crude extract, which warrants further study.
