目的从分子水平证实经太空飞行处理的菜豆种子后代突变系的遗传物质 (DNA)确实发生了基因突变。方法菜豆种子经卫星搭载飞行 1 5d ,回收种植后 ,采用RAPD的方法对 5个叶片形态有变异的突变体的群体及亲本材料进行分析。结果在 50个 1 0寡聚核苷酸引物中 ,有 2 0个引物扩增出了DNA带 ,共有 1 80条带 ,其大小在 2 0 0~ 2 0 0 0bp之间 ,其中有 3个引物的扩增产物与对照相比产生明显的差异。结论本文首次对空间诱变的菜豆品系进行DNA水平上的分析 ,RAPD分析结果说明该突变品系与原始对照在DNA水平上确实已发生了明显变异。
Objective To confirm that genetic mutations have indeed occurred in the genetic material (DNA) of the offspring of offspring of bean seeds treated by space flight at the molecular level. Methods The bean seeds were sown on a satellite for 15 days. After the seeds were planted and recovered, five populations of mutants with variations in leaf shape and their parents were analyzed by RAPD. Results Of the 50 oligonucleotide primers, 20 DNAs were amplified with a total of 180 bands ranging in size from 200 to 200 bp, of which 3 The amplification products of the primers make a significant difference compared to the control. Conclusions This study is the first to analyze the DNA level in space-mutated bean germplasm. The results of RAPD analysis showed that the mutant strain did indeed have significant variation at the DNA level compared with the original control.
