目的 研究低硒 (Se)低维生素 E(VE)能否诱导大鼠肝细胞凋亡及 c AMP和细胞内 Ca2 + 所起的作用。方法 以天然的和人工半合成的低 Se低 VE饲料喂养大鼠 17周 ,采用流式细胞术检测肝细胞凋亡 ,采用放免法检测 c AMP含量 ,Fura- 2负载荧光分光光度法测定细胞内 Ca2 + 含量。结果 与补 Se和 VE组大鼠相比 ,低Se低 VE组大鼠肝细胞凋亡显著增加 ;c AMP含量明显升高 ;细胞内 Ca2 + 含量明显升高。结论 微量营养素 Se和 VE缺乏可诱导大鼠肝细胞凋亡 ;c AMP和细胞内 Ca2 + 可能参与这类细胞凋亡的调控
Objective To investigate the effect of low selenium (Se) vitamin E (VE) on hepatic cell apoptosis and cAMP and intracellular Ca2 + in rats. Methods The natural and artificial semi-synthetic low-Se low-VE feed was used to feed rats for 17 weeks. The apoptosis of hepatocytes was detected by flow cytometry. The content of cAMP was detected by radioimmunoassay. Ca2 + content. Results Compared with rats in Se and VE groups, the apoptosis of hepatocytes in rats with low Se and low VE was significantly increased, the content of cAMP was significantly increased, and the content of intracellular Ca2 + was significantly increased. Conclusion The micronutrient Se and VE deficiency can induce the apoptosis of rat hepatocytes; c AMP and intracellular Ca2 + may be involved in the regulation of apoptosis of these cells
