目的研究不同移植方法对骨骼肌卫星细胞心肌内移植后心肌再生作用的影响。 方法自犬臀部取骨骼肌提取、培养、扩增骨骼肌卫星细胞 ,4’ ,6 -diamidino - 2 - phenylindone(DAPI)荧光标记 ,经局部注射和结扎的左冠状动脉前降支远端灌注移植入自体急性心肌梗塞区域 ,2、4、8周后处死动物 ,取心肌标本行组织形态学检查。结果DAPI标记卫星细胞效果良好 ,在梗塞区、乳头肌及局部注射部位均观察到带荧光的卫星细胞 ,部分已分化成带横纹的肌纤维并以闰盘相连。经冠状动脉灌注移植的卫星细胞排列有序 ,与心肌细胞排列方向一致 ,而局部注射移植的卫星细胞排列紊乱。 结论骨骼肌卫星细胞经冠状动脉灌注移植所形成的肌纤维排列有序 ,分布于整个梗塞区域 ,有利于心肌再生。
Objective To study the effect of different transplantation methods on the myocardial regeneration of skeletal muscle satellite cells after myocardial transplantation. Methods The skeletal muscles from the buttocks of dogs were extracted, cultured and expanded. The skeletal muscle satellite cells were labeled with 4 ’, 6 - diamidino - 2 - phenylindone (DAPI) fluorescent label and then perfused with left anterior descending coronary artery by local injection and ligation Into the acute myocardial infarction area, 2, 4, 8 weeks after the animals were sacrificed, take myocardial samples histological examination. Results DAPI-labeled satellite cells worked well. Fluorescent satellite cells were observed in the infarct area, papillary muscles and local injection sites. Some of them were differentiated into striped muscle fibers and connected with intercalated discs. The satellite cells transplanted by coronary perfusion were arranged in an orderly manner, aligned with the direction of myocardial cells, while the satellite cells transplanted by local injection were disordered. Conclusion Muscle fibers formed by skeletal muscle satellite cells transplanted through perfused coronary arteries are arranged in order and distributed throughout the infarct area, which is beneficial to myocardial regeneration.
