目的探讨汉防己甲素(Tet)及维拉帕米(Ver)抗肝纤维化的机制.方法采用细胞培养、流式细胞术和图像分析法检测Tet及Ver对3T6成纤维细胞增殖的影响.结果不同浓度的Tet及Ver可使3T6细胞G1期或G2+M期细胞显著增多(P<001);Tet(15,20μg)使细胞RNA含量显著下降(P<001);Ver(10,20μg)使细胞DNA含量显著降低(P<001);Tet(15,20μg)和Ver(5,10μg)均使细胞蛋白质含量显著增加(P<001).结论Tet及Ver对成纤维细胞的增殖具有抑制作用,可能是抗肝纤维化的机制之一
Objective To investigate the anti-hepatic fibrosis mechanisms of tetrandrine (Tet) and verapamil (Ver). Methods The effects of Tet and Ver on the proliferation of 3T6 fibroblasts were detected by cell culture, flow cytometry and image analysis. Results Tet and Ver were significantly increased in G1 phase or G2 + M phase of 3T6 cells (P <001). Tet (15, 20 μg) significantly decreased cellular RNA (P <001) ; Ver (10, 20μg) significantly decreased the cellular DNA content (P <001); both Tet (15,20μg) and Ver (5,10μg) increased the cellular protein content significantly 01). Conclusion Tet and Ver have an inhibitory effect on the proliferation of fibroblasts and may be one of the mechanisms of anti-hepatic fibrosis
