目的:探讨改良液氮冷冻法制作非创伤性股骨头缺血坏死(ANFH)的可行性,寻找一种较为理想的ANFH的造模方法。方法:将20只新西兰大白兔利用改良液氮冷冻法,制作ANFH模型,分别在术后2,4,6,8周行大体观、MRI、X线及Masson三色染色观察。结果:造模后2周股骨头变化不明显;造模后4周股骨头表面软骨变性,软骨下骨部分坏死;造模后6周股骨头表面软骨部分剥脱,软骨下骨坏死增多;造模后8周股骨头大部分骨质坏死,甚至出现塌陷。结论:改良液氮冷冻方法制作的股骨头坏死模型,其影像学及组织学改变较为符合早中期股骨头坏死的改变,且此法制作模型简单,费用低,坏死表现早,成功率高。
Objective: To explore the feasibility of making non-traumatic avascular necrosis of femoral head necrosis (ANFH) by modified liquid nitrogen freezing method and to find a better method of modeling ANFH. Methods: ANFH model was made in 20 New Zealand white rabbits by modified liquid nitrogen freezing method. Gross observation, MRI, X-ray and Masson trichrome staining were performed at 2, 4, 6 and 8 weeks after operation. Results: There was no obvious change of femoral head 2 weeks after modeling. Cartilage degeneration and subchondral bone necrosis occurred on the surface of femoral head 4 weeks after model establishment. Partial cartilage exfoliation and subchondral bone necrosis of femoral head surface increased 6 weeks after modeling. After 8 weeks most of the femoral head bone necrosis, or even collapse. Conclusion: The model of femoral head necrosis made by modified liquid nitrogen freezing method is more consistent with the change of early-mid stage osteonecrosis of the femoral head. This method is simple and inexpensive, with early necrosis and high success rate.
