目的对1例入境的美国籍人类免疫缺陷病毒1型(HIV-1)感染者血液样本进行基因序列分析,鉴定其亚型。方法对该名HIV感染者进行流行病学调查,从其血浆样本中提取病毒总RNA,并应用巢式RT-PCR扩增HIV-1gag和pol基因,测序后通过系统进化分析进行亚型鉴定。结果基于gag和pol基因系统进化分析显示,该感染者携带的毒株基因序列均与HIV-1 CRF22_01A1亚型国际参考株聚集成簇,Bootstap值分别为74%和100%,证实其为HIV-1 CRF22_01A1亚型。这是国内首次报道检出HIV-1 CRF22_01A1亚型。结论 HIV-1 CRF22_01A1亚型毒株已随国际旅行者传入我国,应加强对国际旅行人员HIV的分子监测。
OBJECTIVE: To analyze the gene sequence of one HIV-1-infected blood sample of a Chinese immigrant and identify its subtypes. Methods Epidemiological survey of the HIV-infected persons was carried out. The total RNA of the virus was extracted from the plasma samples. The HIV-1 gag and pol genes were amplified by nested RT-PCR and sequenced for the subtype identification by phylogenetic analysis. Results Based on the phylogenetic analysis of gag and pol genes, the gene sequences of the strains carried by this isolate all clustered with the international reference strain of HIV-1 CRF22_01A1 subtype with Bootstap values of 74% and 100%, respectively, confirming that they are HIV- 1 CRF22_01A1 subtype. This is the first report of the detection of HIV-1 CRF22_01A1 subtype. Conclusion HIV-1 CRF22_01A1 subtype strains have been introduced into our country with international travelers, and molecular monitoring of HIV among international travelers should be strengthened.
