探讨P21WAF1/CIP1与p53基因在大肠癌中的表达意义及其关系。方法应用原位杂交及免疫组化技术检测大肠癌中的抑癌基因P21WAF1/CIP1mRNA及p53蛋白的表达。统计学采用卡方检验。结果发现大肠癌组织及癌旁组织中P21WAF1/CIP1表达阳性率分别为84.13%(53/63)及61.90%(39/63),两者之间差别显著(P<0.05)。P21WAF1/CIP1表达程度与癌组织的分化程度有关,两者之间存在显著差别(P<0.05)。P21WAF1/CIP1表达与其他临床病理指标无关(P>0.05)。p53蛋白在大肠癌中阳性率为38.09%(24/63),p53过表达的患者其P21WAF1/CIP1的表达较之p53阴性者为低,二者存在显著差异(P<0.05)。结论P21WAF1/CIP1作为一新的抑癌基因,其表达相当程度上依赖于p53蛋白。
To investigate the expression and significance of P21WAF1/CIP1 and p53 in colorectal cancer. Methods The expression of P21WAF1/CIP1 mRNA and p53 protein in colorectal cancer was detected by in situ hybridization and immunohistochemistry. Chi-square test was used for statistics. The results showed that the positive rate of P21WAF1/CIP1 expression in colorectal cancer tissues and adjacent tissues was 84.13% (53/63) and 61.90% (39/63), respectively. There was a significant difference between them (P<0.05). ). The degree of P21WAF1/CIP1 expression was related to the degree of differentiation of the cancer tissue, and there was a significant difference between the two (P<0.05). The expression of P21WAF1/CIP1 was not related to other clinicopathological parameters (P>0.05). The positive rate of p53 protein in colorectal cancer was 38.09%(24/63). The expression of P21WAF1/CIP1 in patients with p53 overexpression was lower than that in patients with p53 negative. There was a significant difference between them (P<0.05). . Conclusions P21WAF1/CIP1 is a novel tumor suppressor gene whose expression is dependent on p53 protein to a considerable degree.
