目的:考察苦瓜总皂苷对水负荷大鼠的利尿作用及并考察其对大鼠肾功能的影响。方法:筛选排尿量正常的大鼠,并随机分为空白对照组(蒸馏水)、模型对照组(蒸馏水)、阳性对照组(氢氯噻嗪10 mg/kg)、苦瓜总皂苷高剂量组(0.576 g/kg)、中剂量组(0.288 g/kg)、低剂量组(0.144 g/kg),连续8天灌胃给药后,以生理盐水灌胃造成水负荷模型,测定水负荷后4h内的排尿量,并测定尿蛋白浓度,同时测定血肌酐、血尿素氮含量。结果:苦瓜总皂苷0.288 g/kg能显著增加水负荷大鼠在4h内的排尿量,各实验剂量的苦瓜总皂苷均有降低尿蛋白水平的趋势,且Na+、Cl-离子的排泄量增加,但对血肌酐、血尿素氮含量没有明显影响。结论:苦瓜总皂苷(0.288 g/kg)对大鼠有显著的利尿效果,0.144 g/kg~0.576 g/kg的剂量不会引起肾功能的明显改变。
Objective: To investigate the diuretic effect of bitter gourd total saponin on water-loaded rats and its effect on renal function in rats. Methods: The rats with normal urinary volume were screened and divided randomly into blank control group (distilled water), model control group (distilled water), positive control group (hydrochlorothiazide 10 mg / kg), balsam pear total saponin high dose group (0.576 g / kg ), Middle dose (0.288 g / kg) and low dose (0.144 g / kg) for 8 days. The water load model was induced by intragastric administration of saline and the urine output within 4 hours after the water load was measured , And determination of urinary protein concentration, simultaneous determination of serum creatinine, blood urea nitrogen content. Results: Total ginsenoside 0.288 g / kg could significantly increase the amount of urinary excretion in water-loaded rats within 4 h. The total dose of bitter gourd saponin in all experimental groups tended to decrease urinary protein levels, and the excretion of Na + and Cl- ions increased. However, serum creatinine, blood urea nitrogen content did not significantly affect. Conclusion: Total saponin of bitter melon (0.288 g / kg) has significant diuretic effect on rats. The dose of 0.144 g / kg ~ 0.576 g / kg does not cause obvious change of renal function.
