抽提9例成人斑块和斑块旁中膜组织 DNA,部分 DNA 用 HPLC 分析了 DNA 甲基化全貌,另一部分 DNA 用 Msp Ⅰ和 HpaⅡ酶切,用 c-myc 基因探针进行杂交。结果出现:斑块组织 DNA 出现低甲基化;2例斑块组织 DNA中 c-myc 基因部分 CCGG 位点出现脱甲基化。DNA 低甲基化状态可能与基因转录调控有关。
DNA was extracted from 9 adult plaques and plaque adjacent to the medial plaque. Part of the DNA was analyzed by HPLC for DNA methylation. The other DNA was digested with Msp I and Hpa II and hybridized with c-myc gene probe. The results showed that there was hypomethylation of plaque DNA, and demethylation of some CCGG sites of c-myc gene in 2 plaque tissues. DNA hypomethylation may be related to gene transcription regulation.
