目的:探讨Ref-1在正常宫颈组织和宫颈癌中的表达及其生物学意义。方法:采用免疫组织化学SP法和RT-PCR方法检测Ref-1在正常宫颈组织及子宫颈癌组织中的表达。结果:①Ref-1在正常宫颈与高分化宫颈鳞癌组织中的表达差异无统计学意义(P>0.05);随着病理学分级的升高,宫颈鳞癌组织中Ref-1的表达明显降低(P﹤0.01)。②Ref-1 mRNA在正常宫颈及高分化宫颈鳞癌组织中的表达差异无统计学意义(P>0.05),但Ref-1 mRNA在正常宫颈及中、低分化宫颈鳞癌组织中的表达差异有统计学意义(P<0.01)。Ref-1 mRNA的表达随着病理学分级的升高而下降,甚至缺失。结论:Ref-1基因及其编码蛋白在宫颈鳞癌组织中表达随着病理学分级的升高,表达明显下降或缺失,说明Ref-1基因失活与宫颈癌的浸润和转移有关,且Ref-1表达水平与分化程度密切相关。
Objective: To investigate the expression of Ref-1 in normal cervical tissue and cervical cancer and its biological significance. Methods: The expression of Ref-1 in normal cervical tissue and cervical cancer tissues was detected by immunohistochemical SP method and RT-PCR method. Results: ① There was no significant difference in Ref-1 expression between normal cervix and well-differentiated cervical squamous cell carcinoma (P> 0.05). Ref-1 expression in cervical squamous cell carcinoma was significantly decreased with the increase of pathological grade (P <0.01). There was no significant difference in the expression of Ref-1 mRNA between normal cervical tissue and well-differentiated cervical squamous cell carcinoma tissue (P> 0.05), but the expression of Ref-1 mRNA in normal cervical tissue, moderately differentiated and poorly differentiated cervical squamous cell carcinoma Statistical significance (P <0.01). Ref-1 mRNA expression decreased with the increase of pathological grade, or even missing. Conclusion: The expression of Ref-1 gene and its encoded protein in cervical squamous cell carcinoma with pathological grade increased significantly decreased or absent, indicating that Ref-1 gene inactivation and cervical cancer infiltration and metastasis, and Ref -1 expression level is closely related to the degree of differentiation.
