目的:考察乌司他丁干预窒息大鼠心肺复苏后转录因子T-bet和GATA-3的表达。方法:随机将SD大鼠分为3组:假手术组(A组,不进行窒息及心肺复苏,仅进行麻醉和气管切开插管、血管穿刺);对照组(B组,静脉注射生理盐水4mL.kg-1);考察组(C组,乌斯他丁5万U.kg-1,给药容积为4mL.kg-1);采用实验设计模型考察大鼠复苏前后实验设定时间段血清T-bet和GATA-3mRNA表达及白介素-12(IL-12)、白介素-4(IL-4)、r-干扰素(INF-r)、肿瘤坏死因子-α(TNF-α)水平的变化。结果:B、C组复苏后6h分别与复苏后0h及A组比较,IL-12、IFN-γ、TNF-α水平均升高(P<0.01),B组复苏后6hT-bet表达上调,GATA-3表达下调,GATA-3/T-bet下降(P<0.05);与A组比较,C组复苏后6hT-bet表达上调(P>0.05),GATA-3表达上调(P<0.01),GATA-3/T-bet升高(P<0.05);复苏后6hC组与B组同时点比较,IL-12、IFN-γ、TNF-α水平低于B组,GATA-3表达上调、T-bet表达下调、GATA-3/T-bet升高(P<0.01)。结论:心肺复苏可对实验动物GATA-3和T-bet表达造成影响,并使其IL-12、IFN-γ和TNF-α水平升高,乌司他丁介入对此有一定的干预作用。
Objective: To investigate the expression of T-bet and GATA-3 in cardiopulmonary resuscitation rats treated with ulinastatin. Methods: The SD rats were randomly divided into three groups: sham-operation group (group A, no asphyxia and cardiopulmonary resuscitation, anesthesia and endotracheal intubation, vascular puncture only); control group (group B, intravenous saline (C group, Ustattin 50000 U.kg-1, administration volume of 4mL.kg-1); experimental design model was used to study the experimental set time before and after resuscitation Serum levels of T-bet and GATA-3 mRNA and the levels of interleukin-12, IL-4, INF-r and tumor necrosis factor-alpha Variety. Results: The levels of IL-12, IFN-γ and TNF-α in group B and C at 6h after resuscitation were significantly higher than those at 0h and A respectively (P <0.01) GATA-3 / T-bet decreased (P <0.05). Compared with group A, the expression of T-bet increased (P> 0.05) and the expression of GATA-3 increased , GATA-3 / T-bet increased (P <0.05). Compared with B group at 6h after resuscitation, the levels of IL-12, IFN-γ and TNF- T-bet expression was down-regulated and GATA-3 / T-bet was increased (P <0.01). Conclusion: Cardiopulmonary resuscitation may affect the expression of GATA-3 and T-bet in experimental animals and increase the levels of IL-12, IFN-γ and TNF-α. Intervention with ulinastatin may have some intervention effects.
