目的:研究双参宁心方对大鼠心肌缺血/再灌注损伤能量代谢的影响。方法:采用Wistar大鼠结扎冠状动脉左前降支40 min,再灌注2 h,建立心肌缺血/再灌注模型,大鼠随机分为6组:假手术组、模型组、曲美他嗪组(10 mg·kg-1)、双参宁心方低、中、高剂量组(22.5,45,90 mg·kg-1)。预防给药5 d,每日1次,末次给药后1 h,行手术。肌酸激酶同工酶MB测定试剂盒检测血清CK-MB活性;HPLC测定心肌组织ATP,ADP,AMP含量并计算TAN和EC。结果:与模型组比较,双参宁心方预防性给药可降低血清CK-MB活性,同时增加心肌ATP含量和EC水平(P<0.01或P<0.05)。结论:双参宁心方通过保存缺血心肌ATP,维持心肌细胞能荷水平减轻缺血/再灌注损伤。
Objective: To study the effect of Shuangshenningxin Recipe on energy metabolism of myocardial ischemia / reperfusion injury in rats. Methods: The left anterior descending coronary artery was ligated for 40 min and reperfused for 2 h. Wistar rats were randomly divided into 6 groups: sham operation group, model group and trimetazidine group 10 mg · kg-1), low-, medium- and high-dose Shuangshenningxin (22.5, 45, 90 mg · kg-1). Prevention of dosing 5 d, 1 day, 1 h after the last administration, surgery. Creatine kinase MB assay kit serum CK-MB activity; determination of myocardial ATP, ADP, AMP content and calculate TAN and EC. Results: Compared with model group, SHCS could decrease serum CK-MB activity and increase myocardial ATP content and EC (P <0.01 or P <0.05). Conclusion: Shuangshen Ningxin decoction could reduce the ischemia / reperfusion injury by preserving ATP of ischemic myocardium and maintaining the energy level of cardiomyocytes.
