目的研究β-榄香烯对人肝星状细胞(LX-2)Rho信号通路因子GEF、GDIβ表达的影响。方法将对数生长的LX-2细胞培养24h,分别以质量浓度2.5 mg/L,5 mg/L,10 mg/L的β-榄香烯作用LX-2细胞48h后,采用荧光定量PCR和Western blot法分别检测Rho信号通路因子GEF、GDIβmRNA及其蛋白表达水平。结果与空白对照组相比,不同浓度的β-榄香烯作用于肝星状细胞,其GEF mRNA和蛋白水平明显降低,GDIβmRNA和蛋白水平明显升高,差异有统计学意义。结论β-榄香烯可以降低肝星状细胞GEF的表达,上调GDIβ的表达,从而干扰GEF/GDIβ-ROCK通路,发挥抗纤维化的作用。
Objective To study the effect of β-elemene on the expression of Rho signal pathway factors GEF and GDIβ in human hepatic stellate cells (LX-2). Methods LX-2 cells cultured in logarithmic phase were cultured for 24 h. LX-2 cells were treated with 2.5 mg / L, 5 mg / L and 10 mg / L β-elemene respectively for 48 h. Western blot was used to detect Rho signal pathway factors GEF, GDIβmRNA and protein expression levels. Results Compared with the blank control group, the β-elemene concentrations in hepatic stellate cells were significantly decreased, the GEF mRNA and protein levels were significantly decreased, and the mRNA and protein levels of GDIβ were significantly increased. The difference was statistically significant. Conclusion β-elemene can reduce the expression of GEF in hepatic stellate cells and up-regulate the expression of GDIβ, thus interfering with GEF / GDIβ-ROCK pathway and exerting anti-fibrosis effects.
