目的:建立高效液相色谱法测定血浆中佐匹克隆浓度。方法:采用Eurospher 100-5 C_(18)色谱柱(200mm×4.6mm,5μm);流动相:0.02 mol·L~(-1)磷酸二氢钾(含0.2%三乙胺,pH=4.0)-甲醇(38:62);流速:1.0 ml·min~(-1);柱温:30℃;激发波长:315nm,发射波长:385 nm。结果:血浆佐匹克隆在2.0~200.0ng·m1~(-1)浓度范围内线性关系良好(r=0.999 5)。低、中、高3种浓度绝对回收率分别为92.64%,92.97%,94.66%,日内、日间RSD均小于10%。结论:本法可用于佐匹克隆血浆中浓度的测定以及药物动力学研究。
Objective: To establish a method for the determination of zopiclone in plasma by high performance liquid chromatography. Methods: Eurospher 100-5 C 18 column (200 mm × 4.6 mm, 5 μm) was used. The mobile phase consisted of 0.02 mol·L -1 potassium dihydrogen phosphate (containing 0.2% triethylamine and pH = 4.0) - methanol (38:62); flow rate: 1.0 ml · min -1; column temperature: 30 ° C; excitation wavelength: 315 nm, emission wavelength: 385 nm. Results: Zopiclone showed good linearity (r = 0.999 5) within the concentration range of 2.0 ~ 200.0ng · m ~ (-1). The absolute recoveries of low, medium and high concentrations were 92.64%, 92.97% and 94.66%, respectively, and the RSDs were less than 10%. Conclusion: This method can be used for the determination of zopiclone plasma concentration and pharmacokinetic studies.
