目的考察Ⅱ型ZTC1+1天然澄清剂在萝摩多糖提取中的提纯效果。方法以硫酸-苯酚法测定含量,通过正交实验进行优化,确定理想的Ⅱ型ZTC1+1天然澄清剂处理工艺。采用Ⅱ型ZTC1+1天然澄清剂对提取出的萝摩多糖复合物进行纯化得萝摩粗多糖。结果Ⅱ型ZTC1+1天然澄清剂能有效提高萝摩多糖的提取量,并与澄清剂2组份的比例,反应温度有关。结论Ⅱ型ZTC1+l天然澄清剂纯化萝摩总糖的最佳工艺为A3B1C2D1,即A、B组分的体积分数分别为1%、2%,药液的浓缩程度为m(药物/g):V(水/mL)=1:10,温度为70℃,加热时间为B组分加入后加热1 h,A组分加入后加热1 h。
Objective To investigate the purification effect of type Ⅱ ZTC1 + 1 natural clarifying agent on the extraction of polysaccharides from Radix. Methods The content of ZTC1 + 1 natural clarifying agent was determined by sulfuric acid - phenol method and optimized by orthogonal experiment. The crude polysaccharides from Radix Lorola were purified by using natural type Ⅱ ZTC1 + 1 clarifying agent. Results Type Ⅱ ZTC1 + 1 natural clarifying agent can effectively increase the extraction of Lomopolysaccharide, which is related to the proportion of 2 components of clarifying agent and the reaction temperature. Conclusion The best technology of ZTC1 + l natural clarifying agent for purifying total sugar is A3B1C2D1, that is, the volume fraction of A and B is 1% and 2%, respectively, and the concentration of liquid is m (drug / g) : V (water / mL) = 1: 10, the temperature is 70 ℃, the heating time is heating for 1 h after the addition of B component, heating for 1 h after adding A component.
