目的:研究三七总皂苷(PNS)对快速老化SAMP8小鼠大脑α-分泌酶mRNA表达的影响。方法:采用快速老化痴呆模型小鼠SAMP8,将其随机分为对照组、PNS高、低剂量组(200,100 mg.kg-1)和石杉碱甲组(0.3 mg.kg-1),每组8只,对照组给予相同容积的双蒸水灌胃。每天灌胃1次,连续给药2个月。通过实时荧光定量PCR法检测各组SAMP8小鼠大脑α-分泌酶(ADAM9,ADAM10,ADAM17)mRNA的含量。结果:对照组ADAM9 mRNA含量均低于PNS高剂量组和石杉碱甲组(P<0.05);低剂量组ADAM10 mRNA的含量低于对照组(P<0.05);PNS高、低剂量组和石杉碱甲组ADAM17 mRNA与对照组比较无显著差异。结论:PNS可以上调SAMP8小鼠脑内ADAM9 mRNA和下调ADAM10 mRNA的表达。
Objective: To study the effects of Panax Notoginseng Saponins (PNS) on α-secretase mRNA expression in the brain of rapidly aged SAMP8 mice. Methods: SAMP8 was used in the model mice of fast-aging dementia. The rats were randomly divided into control group, PNS high and low dose groups (200, 100 mg.kg-1) and huperzine A group (0.3 mg.kg-1) 8, the control group given the same volume of double distilled water gavage. Gavage 1 day, continuous administration for 2 months. Real-time fluorescent quantitative PCR method was used to detect the content of α-secretase (ADAM9, ADAM10, ADAM17) mRNA in SAMP8 mice. Results: The levels of ADAM9 mRNA in the control group were lower than those in the high-dose PNS and huperzine A groups (P <0.05), while those in the low-dose group were lower than those in the control group (P <0.05) Huperzine A ADAM17 mRNA group compared with the control group no significant difference. Conclusion: PNS can up-regulate the expression of ADAM9 mRNA and down-regulate the expression of ADAM10 mRNA in SAMP8 mice.
