目的深入研究中药复方861抗肝纤维化的机理。方法向传一代肝星状细胞培养液中加入终浓度为10mg/ml的复方861作用48小时,收获细胞培养液观察Ⅰ,Ⅲ,Ⅳ型胶原及TGFβ1的分泌量并测定胶原酶活性;收获肝星状细胞并提取总RNA,斑点杂交法检测胶原、胶原酶及其抑制TIMP1mRNA水平的变化。结果复方861可明显抑制肝星状细胞Ⅰ,Ⅲ,Ⅳ型胶原及TGFβ1mRNA的表达及相应蛋白的分泌,同时可提高间质性胶原酶的活性并抑制TIMP1mRNA的表达。结论复方861在抑制胶原过度合成的同时,又可促进胶原的降解,从而使二者间的失衡得以纠正,达到抗肝纤维化的目的。
Objective To study the mechanism of anti-hepatic fibrosis in Chinese herbal compound 861. METHODS: Compound 861 (10 mg / ml) was added into the culture of primary hepatic stellate cells for 48 hours. The secretions of type Ⅰ, Ⅲ and type Ⅳ collagen and TGFβ1 were harvested and the activity of collagenase was measured. The harvested liver Total RNA was extracted from the astrocytes and the changes of collagen, collagenase and TIMP1 mRNA levels were detected by dot blot hybridization. Results Compound 861 could significantly inhibit the expression of collagen type Ⅰ, Ⅲ, Ⅳ and TGFβ1 mRNA in hepatic stellate cells and the secretion of corresponding proteins, meanwhile, increase the activity of interstitial collagenase and inhibit the expression of TIMP1 mRNA. Conclusion Compound 861 can inhibit the collagen synthesis, but also promote the degradation of collagen, so that the imbalance between the two can be corrected to achieve the purpose of anti-liver fibrosis.
