目的:探讨肥胖老鼠体内心脏、动脉与脂肪组织中骨形态发生蛋白4(BMP4)的表达及其与炎症程度的相关性。方法:将ob/ob小鼠分为实验组与干扰组,以C57BL/6小鼠为对照组。每组各8只,普通饮食喂养,干扰组尾静脉注射BMP4shRNA腺病毒,每周1次,持续12周。12周后心脏采血处死,取心肌组织、腹主动脉及腹部脂肪组织。免疫组织化学染色定位半定量BMP4的表达水平。PCR荧光定量和免疫蛋白印迹法检测BMP4、白细胞介素(IL)-1β和IL-9mRNA与蛋白的表达水平。结果:实验组BMP4、IL-1β和IL-9的mRNA与蛋白在心肌组织和动脉血管的表达均显著高于干扰组和对照组,实验组和干扰组BMP4蛋白在脂肪组织中的表达要显著低于对照组(均P<0.05)。结论:BMP4在肥胖小鼠体内心肌组织和动脉血管中明显高表达,是促进局部炎症反应发生的独立危险因子。
Objective: To investigate the expression of bone morphogenetic protein 4 (BMP4) in heart, artery and adipose tissue in obese mice and its correlation with the degree of inflammation. Methods: Ob / ob mice were divided into experimental group and interference group, C57BL / 6 mice as control group. Each group of eight, fed a normal diet, interference group tail vein injection of BMP4 shRNA adenovirus once a week for 12 weeks. After 12 weeks, the heart blood was sacrificed and the myocardial tissue, abdominal aorta and abdominal adipose tissue were taken. Immunohistochemical staining for semi-quantitative BMP4 expression levels. The expression of BMP4, interleukin (IL) -1β and IL-9 mRNA and protein were detected by PCR and immunoblotting. Results: The mRNA and protein expressions of BMP4, IL-1β and IL-9 in the experimental group were significantly higher than those in the control group and the interference group. The BMP4 protein expression in the experimental group and the interference group was significantly higher than that in the control group Lower than the control group (all P <0.05). Conclusion: BMP4 is overexpressed in myocardium and artery of obese mice and is an independent risk factor for local inflammation.
