目的探讨蜈蚣提取液对裸鼠移植肝癌的治疗作用及机制。方法建立裸鼠Bel-7404人异位肝癌移植模型,以蜈蚣提取液灌胃31d后,用免疫组化法对肿瘤组织标本进行X染色体连锁凋亡抑制蛋白(XIAP)、Bax基因、血管内皮细胞生长因子(VEGF)和促血管生成素2(Ang-2)的检测。结果对照组XIAP,VEGF与Ang-2的染色细胞数、染色强度及OD值均较治疗组多而强,而Bax的染色强度、染色细胞数及OD值较治疗组少而弱,差异均有统计学意义(P<0.01)。结论蜈蚣提取液能抑制裸鼠Bel-7404移植瘤的生长,其机制与抑制肿瘤血管生成、促进肿瘤细胞凋亡有关。
Objective To investigate the therapeutic effect and mechanism of extract of centipede on liver cancer in nude mice. Methods The nude mice model of Bel-7404 human heterotopic liver cancer was established. The centipede extract was administered intragastrically for 31 days. The expression of XIAP, Bax and vascular endothelial cells Detection of growth factors (VEGF) and Angiopoietin 2 (Ang-2). Results The number of staining cells, staining intensity and OD value of XIAP, VEGF and Ang-2 in the control group were more and more than those in the treatment group, while the staining intensity, the number of stained cells and the OD value of Bax in the control group were both less and weaker than those in the treatment group Statistical significance (P <0.01). Conclusion Centipede extract can inhibit the growth of Bel-7404 xenografts in nude mice. Its mechanism is related to inhibiting tumor angiogenesis and promoting tumor cell apoptosis.
