目的研究甲氧化三丁基锡(tributyltin methoxide,TBTMO)通过线粒体途径诱导Jurkat T淋巴瘤细胞凋亡。方法使用不同剂量(0、0.4、0.6、0.8和1.0μmol/L)的TBTMO处理细胞,分别于不同时间点收获细胞后,采用流式细胞仪对磷脂酰丝氨酸(phosphatidylserine,PS)外翻、线粒体Apo2.7蛋白和线粒体膜电位等分子生物学标志和指标进行检测和分析。结果 TBTMO诱导后Jurkat T淋巴瘤细胞的PS外翻,可见有凋亡小体、细胞核凝聚、细胞核碎裂等许多凋亡的典型变化特征。TBTMO诱导后线粒体膜蛋白Apo2.7分子增加、膜电位快速降低并维持这种低电位至濒临死亡。结论 TBTMO可通过线粒体途径诱导Jurkat人淋巴瘤细胞凋亡。
Objective To investigate the apoptosis of Jurkat T lymphoma cells induced by tributyltin methoxide (TBTMO) via the mitochondrial pathway. Methods The cells were treated with TBTMO at different dosages (0, 0.4, 0.6, 0.8 and 1.0 μmol / L), and the cells were harvested at different time points. The effects of phosphatidylserine (PS) Apo2.7 protein and mitochondrial membrane potential and other molecular markers and indicators of detection and analysis. Results After TBTMO induced PS eversion of Jurkat T lymphoma cells, typical apoptotic features such as apoptotic bodies, nuclear condensation and nuclear fragmentation were observed. TBTMO induced mitochondrial membrane protein Apo2.7 molecules increased rapidly decreased membrane potential and maintain this low potential to the brink of death. Conclusion TBTMO induces apoptosis of Jurkat human lymphoma cells through mitochondrial pathway.
