目的:研究乙醇梯度处理和吸附树脂联用对白芍中白芍总苷的纯化工艺,确定最佳工艺条件和参数。方法:采用正交试验法考察乙醇梯度处理对白芍总苷含量的影响,从静态吸附和动态吸附两方面对9种不同型号的大孔吸附树脂进行综合评价,筛选出效果最佳的树脂型号,考察并确定最佳分离纯化条件。结果:最佳醇沉条件为1 g生药/mL的药液用95%乙醇沉至醇浓度为90%,冷藏10 h;以HPD300为最优型号树脂用于分离纯化白芍中白芍总苷,以5倍量50%乙醇洗脱,药材树脂用量比2∶1(2 g药材∶1 mL湿树脂)为最佳工艺。结论:此工艺方法先进合理,适合工业生产,具有简便、快速、准确等优点。
OBJECTIVE: To study the purification process of total glucosides of paeony root in white peony root by the combination of ethanol gradient and adsorption resin, and to determine the optimum technological conditions and parameters. Methods: The effects of ethanol gradient on the content of total glucosides of paeony were studied by orthogonal test. Nine kinds of macroporous adsorption resins were evaluated comprehensively by static adsorption and dynamic adsorption, and the best resin type was screened. Investigate and determine the best separation and purification conditions. Results: The optimal alcohol precipitation condition was that 1 g crude drug / mL solution was precipitated with 95% ethanol to 90% alcohol concentration and refrigerated for 10 h. HPD300 was the best model resin for the isolation and purification of total glucosides of paeony , 5 times the amount of 50% ethanol elution, the amount of resin resin ratio 2: 1 (2 g herbs: 1 mL wet resin) for the best process. Conclusion: The process is advanced and reasonable, suitable for industrial production, with the advantages of simplicity, speed, accuracy and so on.
