目的:对中药天麻中的一种葡聚糖GBⅡ进行研究。方法:天麻药材经过水提醇沉得到天麻粗多糖,经DEAE-Sepharose Fast Flow离子交换色谱柱层析和Sepharose CL-6B纯化得到均一多糖GBⅡ,经13C-NMR、IR谱、单糖组分分析、甲基化分析等方法确定其结构。结果:GBⅡ相对分子量为4300,由27个葡萄糖组成的葡聚糖,以α-D-1,4连接的葡萄糖构成主链,在葡萄糖6位带有少量分支;其中尚含有极少量葡萄糖醛酸。结论:GBⅡ为从天麻中首次分离得到的均一多糖。
Objective: To study a kind of dextran GBⅡ in Chinese traditional medicine Tianma. Methods: Gastrodia elata Blume polysaccharides were obtained through water precipitation and alcohol precipitation, purified by DEAE-Sepharose Fast Flow ion exchange chromatography and purified by Sepharose CL-6B to get homogenous polysaccharides GBⅡ. The 13C-NMR, IR spectra, monosaccharide composition analysis , Methylation analysis and other methods to determine its structure. Results: The relative molecular mass of GBⅡ was 4300. The dextran composed of 27 glucose, the main chain of glucose linked by α-D-1,4, had a few branches in glucose 6, and contained very little glucuronic acid . Conclusion: GB Ⅱ is the first polysaccharide isolated from Gastrodia elata.
