为建立一个应用McAbs测定人血清总IgE的ELISA系统,我们在前文中对6个抗人IgE McAbs及其不同组合方式进行了比较和筛选,决定采用两个针对不同抗原决定簇的McAbs分别作为固相抗体和结合物的方案并确定了McAb试剂的最适工作条件。本文报告我们对McAb-和PcAb-ELISA系统的相关性进行测定并对造成两者测定值差异的原因进行实验分析的结果。
To establish an ELISA system for the determination of total human serum IgE using McAbs, we compared and screened six anti-human IgE McAbs and their different combinations in the previous section and decided to use two McAbs against different epitopes as solid Phase antibody and conjugate protocols and to determine the optimal conditions McAb reagents. This paper reports the results of our experimental analysis of the correlation between McAb- and PcAb-ELISA systems and the reasons for the differences between the two measured values.
