对前期筛选获得和在田间试验中防治水稻细菌性条斑病效果较好的解淀粉芽胞杆菌Bacillusamyloliquefaciens Lx-11,通过Plackett-Burman试验设计及Box-Behnken设计的响应曲面法(response surfacemethodology,RSM)对影响生防菌株Lx-11发酵培养基和培养条件进行了优化。培养基3个主要因子的质量浓度分别为玉米淀粉11.2 g.L 1、黄豆粉14.6 g.L 1和酵母膏0.62 g.L 1。培养条件优化试验得出:培养温度、通气量和接种量是影响发酵活菌数的主要因子,由所得响应曲面方程预测出这3个主要因子分别为30.84°C、59.68 mL/250 mL和1.52%时,在初始pH为7.0,150 r.min 1培养48 h的条件下,发酵活菌数和抑菌带宽的最大值分别为3.75×109cfu.mL 1和8.2 mm。经摇瓶发酵试验和抑菌活性验证该理论预测值与实际值无显著差异。
