目的检测阵发性睡眠性血红蛋白尿(PNH)与再生障碍性贫血(AA)患者外周血血浆中造血负性调控因子IFN-γ、TNF-α和TGF-β1的表达;研究IFN-γ、TNF-α和TGF-β1表达水平与PNH和AA之间的关系。方法应用ELISA法检测18例成人PNH与20例AA患者外周血血浆中IFN-γ、TNF-α和TGF-β1的表达,并与15例健康体检者对比。结果AA组的IFN-γ和TNF-α均高于对照组,P<0.05;TGF-β1显著高于对照组,P<0.01。PNH组的IFN-γ、TNF-α和TGF-β1均显著高于对照组,P<0.01。PNH组的TNF-α高于AA组,P<0.05;PNH组IFN-γ和TGF-β1均显著高于AA组,P<0.01。结论IFN-γ、TNF-α和TGF-β1作为造血负性调控因子在PNH与AA患者体内对造血细胞起抑制作用,反映造血调控因子分泌紊乱可能在PNH与AA的发病中起一定作用。
Objective To detect the expression of IFN-γ, TNF-α and TGF-β1 in peripheral blood of patients with paroxysmal nocturnal hemoglobinuria (PNH) and aplastic anemia (AA) α and TGF-β1 expression levels and the relationship between PNH and AA. Methods The expression of IFN-γ, TNF-α and TGF-β1 in peripheral blood of 18 patients with PNH and 20 patients with AA were detected by ELISA and compared with 15 healthy controls. Results The levels of IFN-γ and TNF-α in AA group were significantly higher than those in control group (P <0.05). The levels of TGF-β1 in AA group were significantly higher than those in control group (P <0.01). The levels of IFN-γ, TNF-α and TGF-β1 in PNH group were significantly higher than those in control group, P <0.01. The level of TNF-α in PNH group was higher than that in AA group, P <0.05. The levels of IFN-γ and TGF-β1 in PNH group were significantly higher than those in AA group (P <0.01). Conclusions IFN-γ, TNF-α and TGF-β1 as hematopoietic negative regulator play an inhibitory role in hematopoietic cells in patients with PNH and AA, indicating that disorder of hematopoietic regulation factors may play a role in the pathogenesis of PNH and AA.
