背景:前期实验表明,作者自制的中药复方安胎养血合剂能够抑制肾移植大鼠急性排斥反应,延长受体存活时间。目的:观察安胎养血合剂对异基因肾移植大鼠移植肾肾小管上皮细胞凋亡及Fas/FasL基因表达的影响,并与同基因肾移植大鼠进行对比。设计、时间及地点:随机对照动物实验,于2005-07/2006-06在辽宁医学院附属第一医院外科实验室完成。材料:选用雄性SD大鼠24只,雄性Wistar大鼠48只。方法:按体质量相近配对后,按随机数字表法分为3组,每组12对。对照组大鼠同基因肾移植(Wistar→Wistar大鼠),生理盐水灌胃;生理盐水组大鼠异种基因肾移植(SD→Wistar大鼠),肾移植后不用任何免疫抑制剂,生理盐水灌胃;安胎养血合剂组大鼠异种基因肾移植(SD→Wistar大鼠),肾移植后给安胎养血合剂灌胃,灌胃剂量均为10mL/(kg·d),移植后1次/d。主要观察指标:于移植后第5天处死动物取肾脏组织,通过原位末端标记法检测凋亡指数,采用反转录-聚合酶链反应检测Fas/FasLmRNA表达。结果:生理盐水组移植肾组织凋亡细胞最多,对照组凋亡细胞极少见。生理盐水组和安胎养血合剂组凋亡指数显著高于对照组(P<0.05),安胎养血合剂组凋亡指数显著低于生理盐水组(P<0.05)。各组中均有FasmRNA的表达,FasLmRNA的表达水平在安胎养血合剂组中明显低于生理盐水组(P<0.05)。结论:安胎养血合剂可以通过下调大鼠移植肾肾小管上皮细胞FasL的mRNA表达,从而减少移植肾的细胞凋亡,起到免疫抑制剂的作用。
BACKGROUND: Previous experiments showed that the author’s self-made Chinese herbal compound Anzi Yangxue mixture can inhibit acute rejection in kidney transplanted rats and prolong the survival time of the recipients. OBJECTIVE: To observe the effect of Anyu Yangxue mixture on renal tubular epithelial cell apoptosis and Fas/FasL gene expression in allogeneic renal allograft rats, and to compare it with syngeneic renal allograft rats. DESIGN, TIME AND SETTING: The randomized controlled animal experiment was performed at the Surgical Laboratory of the First Affiliated Hospital of Liaoning Medical College from July 2005 to June 2006. MATERIALS: Twenty-four male SD rats and 48 male Wistar rats were used. Methods: According to the similar pairing of body mass, they were divided into 3 groups according to the random number table method, with 12 pairs in each group. Rats in the control group were treated with syngeneic kidney transplantation (Wistar→Wistar rats) and normal saline. Rats in the normal saline group were transplanted with xenogeneic genes (SD→Wistar rats). There was no immunosuppressive agent after renal transplantation. Stomach; Anzi Yangxue Mixture group Rat gene xenogeneic kidney transplantation (SD → Wistar rats), kidney transplantation to Antuo Yangxue mixture gavage, intragastric doses were 10mL / (kg · d), after transplantation 1 Times/d. MAIN OUTCOME MEASURES: The kidneys were sacrificed on the fifth day after transplantation. The apoptosis index was detected by in situ end labeling and the expression of Fas/FasL mRNA was detected by reverse transcription-polymerase chain reaction. RESULTS: Apoptotic cells were most abundant in the renal tissue of the saline group, while apoptotic cells were rare in the control group. The apoptotic index of saline group and Antai Yangxue mixture group was significantly higher than that of the control group (P<0.05). The anorexia index of Anfu Yangxue mixture group was significantly lower than that of the saline group (P<0.05). Fas mRNA expression was observed in all groups, and the expression level of FasL mRNA was significantly lower in the Antonyangxue mixture group than in the saline group (P<0.05). Conclusion: Anfuyangxue Mixture can down-regulate the expression of FasL mRNA in renal tubular epithelial cells of rat transplanted kidneys, thus reducing the apoptosis of transplanted kidneys and acting as an immunosuppressive agent.
