目的:探讨桑根中提取分离的2,5′-二羟基-4,3′-二(β-D-吡喃糖氧基)-反式二苯乙烯(DGTS)对HepG2肝癌细胞的抑制增殖和诱导凋亡的作用。方法:应用台盼蓝排斥法检测不同质量浓度DGTS对HepG2细胞的增殖抑制作用;琼脂糖凝胶电泳测定细胞DNA的片段化;应用流式细胞仪检测细胞凋亡率。结果:DGTS作用72 h后,HepG2细胞的增殖明显受到抑制。不同浓度DGTS作用48 h后,出现HepG2细胞凋亡特征性的DNA梯度降解。经DGTS作用24、48、72 h后,HepG2细胞的凋亡率分别为0.96%、2.05%、3.29%。结论:DGTS能明显抑制HepG2细胞增殖,并存在剂量-效应关系和时间-效应关系。
Objective: To investigate the inhibitory and inhibitory effects of 2,5′-dihydroxy-4,3′-di(β-D-pyranoyoxy)-transstilbene (DGTS) extracted and extracted from mulberry root on HepG2 hepatoma cells. And induce apoptosis. Methods: The trypan blue exclusion method was used to detect the proliferation inhibition of HepG2 cells with different concentrations of DGTS. The DNA fragmentation was detected by agarose gel electrophoresis. The apoptosis rate was detected by flow cytometry. RESULTS: After 72 hours of DGTS treatment, the proliferation of HepG2 cells was significantly inhibited. After 48 h of treatment with different concentrations of DGTS, there was a characteristic DNA gradient degradation of HepG2 cells. After 24, 48 and 72 hours of DGTS treatment, the apoptosis rates of HepG2 cells were 0.96%, 2.05%, and 3.29%, respectively. Conclusion: DGTS can significantly inhibit HepG2 cell proliferation, and there is a dose-effect relationship and time-effect relationship.
