目的:观察四物降脂颗粒对过氧化氢(H2O2)致人脐静脉内皮细胞(HUVECs)损伤的保护作用。方法:制备四物降脂颗粒大鼠含药血清,随机分为4组:空白组,H2O2模型组,药物组,丹参素对照组。倒置显微镜下观察细胞形态、密度;噻唑蓝(MTT)法检测细胞活力;比色法测定细胞培养液中超氧化物歧化酶(SOD)活性及细胞内丙二醛(MDA)含量;RT-PCR方法测定细胞内皮素1(ET-1),血栓调节蛋白(TM)mRNA的表达。结果:四物降脂颗粒含药血清可抑制过氧化氢所致的细胞活力降低,减少MDA生成,提高SOD活性;并可抑制ET-1mRNA的表达,升高TMmRNA的表达。结论:四物降脂颗粒含药血清对过氧化氢所致HUVECs损伤具有明显的保护作用。
Objective: To observe the protective effect of Siwu Jiangzhi Granules on the injury of human umbilical vein endothelial cells (HUVECs) induced by hydrogen peroxide (H2O2). Methods: Four serum-containing lipid-lowering rats were prepared and randomly divided into 4 groups: blank group, H2O2 model group, drug group and Danshensu control group. Cell morphology and density were observed under inverted microscope. Cell viability was assayed by MTT assay. SOD activity and MDA content in cell culture medium were determined by colorimetric assay. RT-PCR The expression of endothelin 1 (ET-1) and thrombomodulin (TM) mRNA was measured. Results: Siwu Jiangzhi Granule could inhibit the decrease of cell viability induced by hydrogen peroxide, decrease the production of MDA and increase the activity of SOD. It also inhibited the expression of ET-1 mRNA and increased the expression of TMmRNA. CONCLUSION: Siwu Jiangzhi Capsule-containing serum has obvious protective effect on HUVECs induced by hydrogen peroxide.
