黄瓜绿斑驳花叶病毒(CGMMV)在世界范围内对葫芦科作物造成严重危害,其危害症状因寄主而异,包括严重花叶、变色及畸形。运用RT-PCR技术,对黄瓜绿斑驳花叶病毒进行分子鉴定,并与其他CGMMV分离物进行系统发育分析。同时,以14个不同的葫芦科品种为试材,在苗期人工摩擦接种CGMMV,通过Dot-ELISA技术对接毒幼苗进行带毒率检测。结果表明,本实验(浙江杭州)CGMMV分离物与我国其他省份(海南、广东、辽宁、安徽)及韩国的分离物亲缘关系较近,相似度为99.9%~100%,与来自中国浙江杭州地区(建德、萧山)、江苏、山东的分离物相似度为99.5%~99.6%,与俄罗斯、美国的分离物亲缘关系较远,相似度在95.3%~95.4%之间,表明本实验分离物可能与同源性较高的其他地区的分离物具有相同的侵染来源。抗性评价结果显示,在西瓜的9个品种中,浙蜜二号、丰乐五号为中度抗病,而最易感病的品种是抗病京欣,为中度感病;而在其他瓜类品种中,黄瓜津优一号和津绿一号为中度抗病;蒲瓜浙蒲六号和青蒲一号分别表现为低度感病和中度感病;瓠瓜圆瓠一号表现为低度感病。
Cucumber green mottle mosaic virus (CGMMV) causes serious damage to Cucurbitaceae crops around the world. Its harmful symptoms vary from host to host, including severe mosaic, discoloration and deformity. The molecular identification of Cucumber green mottle mosaic virus was carried out by RT-PCR and phylogenetic analysis was performed with other CGMMV isolates. In the meantime, 14 different Cucurbitaceae cultivars were used as experimental material to inoculate CGMMV by artificial rubbing at the seedling stage, and the poisoning rate of the seedlings was detected by Dot-ELISA. The results showed that the CGMMV isolates in our experiment (Hangzhou, Zhejiang Province) had close genetic relationship with isolates from other provinces in China (Hainan, Guangdong, Liaoning, Anhui) and Korea. The similarities were between 99.9% and 100% (Jiande, Xiaoshan), Jiangsu, Shandong isolates similarity 99.5% ~ 99.6%, and Russia, the United States isolates distant genetic similarities between 95.3% ~ 95.4%, indicating that the experimental isolates May have the same source of infection as isolates from other areas of higher homology. Resistance evaluation results showed that among the 9 varieties of watermelon, Zhemi 2 and Fengle 5 were moderately resistant to disease while the most susceptible varieties were resistant to Jingxin and moderately susceptible; Among the other melon varieties, the cucumber Jinyou No.1 and Jinlv No.1 were moderately resistant; the melon Qinpu 6 and Qingpu 1 showed low and moderate susceptibility respectively; No. Low grade symptoms.
