目的:为探讨IL2基因导入肝癌浸润淋巴细胞及对其生物学活性的影响。方法:应用逆转录病毒载体介导IL2基因导入人肝癌浸润淋巴细胞(TIL),同时对经转导IL2基因的肝癌浸润淋巴细胞的杀伤活性,增殖能力及表型等进行了观察。结果:转导IL2基因后的肝癌浸润淋巴细胞能表达一定量的IL2,明显高于未转导细胞,但其杀伤肿瘤细胞活性,增殖能力及表型等没有显著变化。结论:应用逆转录病毒载体介导IL2基因导入人肝癌浸润淋巴细胞获得成功,亦有IL2基因产物表达。
Objective: To investigate the effect of IL2 gene infiltrating hepatoma infiltrating lymphocytes and their biological activities. METHODS: The retroviral vector-mediated IL2 gene was introduced into human hepatoma infiltrating lymphocytes (TIL). At the same time, the killing activity, proliferative capacity and phenotype of hepatoma infiltrating lymphocytes transduced with IL2 gene were observed. . Results: The infiltrating lymphocytes of hepatoma after transduction of IL-2 gene could express a certain amount of IL-2, which was significantly higher than that of non-transduced cells, but its activity, proliferation ability and phenotype of tumor cells were not significantly changed. Conclusion: The retroviral vector-mediated IL2 gene was successfully introduced into human hepatoma infiltrating lymphocytes, and IL2 gene products were also expressed.
