自1906年Harrison创建细胞培养技术以来,在各学科领域内得到了广泛应用,尤其近年来在毒理学中的应用更引人注目,已逐渐形成了一门新的学科-细胞毒理学(cytotoxicology) 一、细胞培养 将离体细胞在体外(in vitro)模拟体内环境维持其代谢与生存,进行传代与培养称为细胞培养(cell culture)。在细胞毒理研究中可用原代、二倍体细胞及生物学特性明确的细胞株等。 (一)离体细胞于体外存活的适宜条件 1.温度:最适宜的温度为37(36~38)℃。 2.气体与氢离子浓度:O_2可供给细胞呼吸,CO_2为代谢产物并维持培养液的缓冲能力。适于细胞生存的pH为7.2~7.4。
