采用杂交瘤技术制备单克隆抗体中的关键步骤之一是建立一个敏感度高、特异性强、简便快速的筛选方法,以确定需进一步克隆化的杂交瘤细胞。ELISA由于将抗原抗体特异性反应与酶的高度催化作用有机地结合起来,是一个比较理想的方法。用该法检测针对胃癌细胞株(MGC80·3)的单克隆抗体,结果满意。
One of the key steps in the preparation of monoclonal antibodies using hybridoma technology is to establish a sensitive, specific, simple and rapid screening method for identifying hybridoma cells that require further cloning. ELISA is an ideal method because of the organic combination of the antigen-antibody specific reaction and the high catalytic activity of the enzyme. This method was used to detect monoclonal antibodies against gastric cancer cell lines (MGC80.3) and the results were satisfactory.
