目的:分析不同年龄组包皮组织中表皮干细胞Integrinβ1的表达,为组织工程学种子细胞的选用提供依据。方法:利用DispaseⅡ和BSA共同作用,分散表皮和真皮组织部分。对不同年龄组分离的表皮干细胞进行Integrinβ1免疫荧光染色,利用流式细胞仪检测不同年龄组Integrinβ1表达的阳性率。结果:不同年龄组分离得到的表皮干细胞形态一致。细胞生长状态和生长速度以7岁年龄组为最佳,其次是16岁年龄组,最后是30岁年龄组。流式细胞仪检测不同年龄组Integrinβ1的表达,7岁组阳性率为93%,16岁组阳性率为56%,30岁组阳性率仅为3.5%。结论:随着年龄的增长,皮肤组织中表皮干细胞的含量降低,低年龄段皮肤组织是组织工程学种子细胞的良好选择。
OBJECTIVE: To analyze the expression of integrinβ1 in the foreskin stem cells of different age groups and provide basis for selection of tissue engineering seed cells. Methods: Dispase Ⅱ and BSA act together to disperse the epidermal and dermal tissue fractions. Epidermal stem cells isolated from different age groups were immunostained by Integrinβ1, and the positive rate of Integrinβ1 expression in different age groups was detected by flow cytometry. Results: The epidermal stem cells isolated from different age groups had the same morphology. The cell growth status and growth rate were best in the 7-year-old group, followed by the 16-year-old group, and finally in the 30-year-old group. The expression of Integrinβ1 in different age groups was detected by flow cytometry. The positive rate was 93% in 7-year-old group, 56% in 16-year-old group, and 3.5% in 30-year-old group. Conclusion: With the increase of age, the content of epidermal stem cells in skin tissue decreases. Skin tissue of lower age is a good choice for tissue engineering seed cells.
