目的:研究紫花牡荆素(casticin,CAS)是否抑制卵巢癌OVCAR-3细胞系肿瘤球形成能力。方法:肿瘤球形成法检测不同浓度CAS(1.0、3.0、10.0μmol/L)对OVCAR-3细胞系肿瘤球形成能力的影响。Western Blot分析探讨CAS影响OVCAR-3细胞系肿瘤球形成机制是否涉及其下调干细胞标志物CD44蛋白表达。结果:CAS以浓度依赖方式抑制OVCAR-3细胞系肿瘤球形成能力。与亲本细胞相比,OVCAR-3细胞系球形成细胞高表达CD44蛋白。CAS显著下调OVCAR-3细胞系球形成细胞CD44蛋白表达。结论:CAS抑制卵巢癌OVCAR-3细胞系肿瘤球形成与其下调干细胞标志物CD44蛋白表达相关。
OBJECTIVE: To investigate whether casticin (CAS) inhibits the tumor formation ability of ovarian cancer OVCAR-3 cells. Methods: Tumor spheroid formation assay was used to determine the effect of different concentrations of CAS (1.0, 3.0 and 10.0 μmol / L) on the tumorigenicity of OVCAR-3 cells. Western Blot analysis to explore whether CAS affects OVCAR-3 cell tumorigenesis is involved in its downregulation of stem cell marker CD44 protein expression. Results: CAS inhibited the tumor formation ability of OVCAR-3 cells in a concentration-dependent manner. OVCAR-3 cell line spheroplasts overexpress CD44 protein compared to parental cells. CAS significantly down-regulated CD44 protein expression in OVCAR-3 cell line. CONCLUSIONS: CAS inhibits tumor spheroid formation in ovarian cancer OVCAR-3 cell lines in relation to its downregulation of stem cell marker CD44 protein expression.
