目的:本研究旨在观察气道高反应大鼠肺组织AQP5的分布和表达。方法:取14只SPF级Wister大鼠随机分为2组:正常对照组;臭氧应激组;每组7只。运用Western Blot,RT-PCR和原位杂交的方法,检测大鼠肺组织AQP5的表达量,定位AQP5在肺内的分布。结果:臭氧应激组大鼠AQP5主要分布在肺泡毛细血管内皮细胞和肺泡Ⅰ型上皮细胞,其表达量明显低于正常对照组。结论:在气道高反应过程中,AQP5的表达下调可能与气道失稳态有关。
Aims: The aim of this study was to observe the distribution and expression of AQP5 in lung tissue of rats with airway hyperresponsiveness. Methods: Fourteen SPF Wister rats were randomly divided into 2 groups: normal control group, ozone stress group, 7 rats in each group. Western Blot, RT-PCR and in situ hybridization methods were used to detect the expression of AQP5 in the lung tissue of rats, and to locate the distribution of AQP5 in the lung. Results: AQP5 was mainly distributed in alveolar capillary endothelial cells and alveolar type I epithelial cells in ozone-stressed rats, and its expression was significantly lower than that in normal control rats. Conclusion: In the process of airway hyperresponsiveness, the down-regulation of AQP5 may be related to the airway destabilization.
