利用原代细胞培养技术,观察了不同浓度的铅(Pb)对支持细胞(Sertoli)功能的影响。结果发现,Pb2+(10-7~10-4mol/L)使处理24h的Sertoli细胞乳酸含量显著升高;Pb2+(10-5~10-4mol/L)也使丙酮酸含量明显升高。Pb2+(10-5mol/L)使处理6h和24h的Sertoli细胞乳酸含量分别显著低于和高于对照水平;Pb2+(10-7~10-4mol/L)使处理24h的Sertoli细胞内cAMP含量显著下降。醋酸锌(Zn2+)对培养的Sertoli细胞的上述观察指标均无影响。因此,用这些指标的变化可以评价Pb2+对睾丸Sertoli细胞的某些毒性。
Using primary cell culture techniques, the effect of different concentrations of lead (Pb) on the function of Sertoli cells was observed. The results showed that Pb2 + (10-7 ~ 10-4mol / L) significantly increased the content of lactic acid in Sertoli cells treated with Pb2 + (10-5 ~ 10-4mol / L). Pb2 + (10-5mol / L) significantly decreased the lactic acid content of Sertoli cells treated for 6h and 24h, respectively. The level of cAMP in Sertoli cells treated with Pb2 + (10-7-10-4mol / L) for 24h was significantly higher than that of control decline. Zinc acetate (Zn2 +) had no effect on the above observations of cultured Sertoli cells. Therefore, some of the toxicity of Pb2 + to testicular Sertoli cells can be assessed using changes in these indicators.
