目的 了解幽门螺杆菌( Hp) 感染对胃粘膜Cmyc 基因蛋白和表皮生长因子受体(EGFR) 表达的影响.方法 应用流式细胞术和免疫荧光染色技术,对Hp 清除前后胃粘膜Cmyc 基因蛋白和EGFR 进行定量测定,用免疫组化法研究增殖细胞核抗原(PCNA) 在组织中的表达.结果 Hp 阳性患者胃粘膜细胞Cmyc 蛋白和EGFR 的阳性细胞百分率和平均荧光强度显著高于 Hp 阴性患者,经治疗Hp转阴者上述指标显著下降,而Hp 仍为阳性者上述指标则无明显变化. 胃粘膜PCNA 标记指数与Cmyc 蛋白和EGFR 呈一致性改变.结论 Hp 感染导致的Cmyc 蛋白和EGFR 的过度表达,可能对细胞增殖和癌变具有促进作用.
Objective To understand the effect of Helicobacter pylori (Hp) infection on the expression of Cmyc gene protein and epidermal growth factor receptor (EGFR) in gastric mucosa. Methods Flow cytometry and immunofluorescence staining techniques were used to quantitatively measure the Cmyc gene protein and EGFR in gastric mucosa before and after Hp removal. Immunohistochemistry was used to study the expression of proliferating cell nuclear antigen (PCNA) in tissues. Results The percentage of C-myc protein and EGFR positive cells and the mean fluorescence intensity of gastric mucosal cells in Hp-positive patients were significantly higher than those in Hp-negative patients. The above-mentioned indexes of Hp-negative patients were significantly decreased in Hp-positive patients, while those in Hp-positive patients were not significant. Variety. The PCNA labeling index of gastric mucosa was consistent with that of Cmyc protein and EGFR. Conclusion Overexpression of Cmyc protein and EGFR caused by Hp infection may promote cell proliferation and canceration.
