目的:探讨葛根成分3’-甲氧基葛根素(3’-methoxy-puerarin,3’-mo-pue)对脑缺血及血流变的影响。方法:将动物随机分为对照组、模型组、葛根素(puerarin,Pue)组和3’-mo-pue组,采用三氯化铁覆盖大脑中动脉造成大鼠局灶性脑缺血模型(MCAO),观察其对脑缺血的影响;采用皮下注射角叉菜胶10mg/kg造成体内血栓模型,进行血流变分析。结果:3’-mo-pue可显著降低MCAO模型大鼠脑梗死范围;明显降低体内血栓模型大鼠尾部血栓长度、全血粘度、血浆粘度、血小板聚集率,减少纤维蛋白含量。结论:3’-mo-pue具有脑保护作用,其机制可能与改善其血液流变学有关。
Objective: To investigate the effects of 3’-methoxy-puerarin (3’-methoxy-puerarin) on cerebral ischemia and hemorrheology. Methods: The animals were randomly divided into control group, model group, puerarin (Pue) group and 3’-mo-pue group. The rats were subjected to focal cerebral ischemia with ferric chloride MCAO) to observe its impact on cerebral ischemia; subcutaneous injection of carrageenan 10mg / kg in vivo thrombosis model for hemorheological analysis. Results: 3’-mo-pue significantly reduced the range of cerebral infarction in rats with MCAO; significantly reduced the tail thrombus length, whole blood viscosity, plasma viscosity, platelet aggregation rate and fibrin content in in vivo thrombus model rats. CONCLUSION: 3’-mo-pue has neuroprotective effect and its mechanism may be related to the improvement of hemorheology.
