应用不同类型的表面活性剂系统研究其对大鼠肝癌GD_3合成酶(CMP-唾液酸-GM_3唾液酸转移酶,ST2)活性的影响及增溶作用。研究结果表明Triton系列的表面活性剂增溶作用较好,在0.1mol/L KCl存在下Tritoa CF-54和Triton X-100(1:1V/V)两者混溶时,其增溶效果比其单独使用更佳,非离子型表面活性剂NP-40和Triton CF-54具有最大的激活作用。当离子型表面活性剂SDS的胆酸钠浓度在0.1%~0.2%时对ST2略有激活作用,面大于0.4%时有抑制作用。不同的表面活性剂对SF2活性产生不同的影响,提示SF2与脂结合的结构区域,与表面活性剂或脂结合时对ST2活性具有调节作用。
Different types of surfactant systems were used to study the effect of solubilizing the GD_3 synthase (CMP-sialic acid-GM-3, sialyltransferase, ST2) in rat liver cancer and solubilization. The results showed that Triton series surfactant had better solubilization effect. When the mixture of Tritoa CF-54 and Triton X-100 (1: 1 V / V) was dissolved in the presence of 0.1 mol / L KCl, It is better to be used alone with the largest activation of the non-ionic surfactants NP-40 and Triton CF-54. When the concentration of sodium cholate of ionic surfactant SDS is between 0.1% and 0.2%, ST2 is slightly activated. When the surface area is larger than 0.4%, the inhibitory effect is observed. Different surfactants have different effects on SF2 activity, suggesting that SF2 binds to lipid structural domains and regulates ST2 activity when combined with surfactants or lipids.
