目的探究子宫内膜异位症(内异症)的发生发展与线粒体能量代谢的关系。方法收集20例内异症患者的在位内膜、盆腔子宫内膜异位灶和卵巢子宫内膜异位囊肿壁组织为异位症组,20例非内异症患者的在位内膜作为对照组,根据差速离心法提取线粒体DNA,采用实时荧光定量PCR扩增方法来检测线粒体DNA中细胞色素B(Cyt B)和烟酰胺腺嘌呤二核苷二钠(NADH)的表达,并比较不同分期、年龄及疼痛程度对Cyt B和NADH表达的影响。结果异位症组患者异位内膜Cyt B和NADH的表达高于对照组、异位症在位内膜和异位囊肿组(P<0.05),内异症Ⅰ、Ⅱ期和Ⅲ、Ⅳ期患者异位内膜的Cyt B和NADH表达高于对照组(P<0.05),不同年龄段异位内膜患者CytB和NADH的表达均高于对照组(P<0.05)。不同疼痛程度患者异位内膜CytB和NADH的表达均高于对照组(P<0.05)。结论线粒体能量代谢可能与内异症的发生发展相关,为内异症的演变提供了生物能量。
Objective To investigate the relationship between the occurrence and development of endometriosis (endometriosis) and mitochondrial energy metabolism. Methods Twenty patients with endometriosis, pelvic endometriosis and ovarian endometriosis were collected from 20 patients with endometriosis. The eutopic endometrium of 20 patients with non-endometriosis In the control group, mitochondrial DNA was extracted by differential centrifugation and the expression of cytochrome B (Cyt B) and nicotinamide adenine dinucleoside disodium (NADH) in mitochondrial DNA was detected by real-time fluorescent quantitative PCR. Effect of different stages, age and degree of pain on Cyt B and NADH expression. Results The expression of Cyt B and NADH in ectopic endometrium of ectopic group was higher than that of control group, ectopic endometrium and ectopic cyst group (P <0.05), endometriosis stage Ⅰ, Ⅱ and Ⅲ, Ⅳ The expression of Cyt B and NADH in ectopic endometrium of patients with ectopic endometrium was higher than that of the control group (P <0.05). The expression of CytB and NADH in ectopic endometrium of different age groups was higher than that of the control group (P <0.05). The expression of CytB and NADH in ectopic endometrium of patients with different pain degree were higher than that of the control group (P <0.05). Conclusion Mitochondrial energy metabolism may be related to the occurrence and development of endometriosis, which provides bioenergetic energy for the evolution of endometriosis.
